Gibson Assembly® RapidAMP™ reagents are available in a benchtop reagent kit or in automated format, compatible with the BioXp™ 3200 Gene Builder. The BioXp™ system enables up to 24 constructs to be built, cloned, and amplified in a single overnight run, eliminating the need to transform into E. coli.
The benchtop RapidAMP™ process begins with a linearized vector plus up to three linear DNA insert fragments, 300-1800 bp each. All are assembled via Gibson Assembly® technology to generate a circular cloning product. This workflow is expanded, automated, and scaled-up using the BioXp™ 3200 Gene Builder. The BioXp™ system starts by synthesizing the required fragments, up to 24 at a time, and then clones them into the vectors of interest (up to 4) in an overnight run, on the instrument. By the following afternoon, these clones will have been amplified to > 10 µg DNA using Gibson Assembly® RapidAMP™ technology. Only with BioXp™ system is it possible to shorten the design-build-test cycle to just 2 days.
The RapidAMP™ workflow produces clean, high-yield DNA material. A representative gel shows clean bands post-restriction digest and before any DNA purification. After cloning and amplification on the bench, or with the BioXp Gene Builder, the final DNA product can be digested to linear form and is then ready for downstream applications, negating the need for traditional E. coli transformation and plasmid purification. RapidAMP™ has been used successfully for transfection (lipofection and electroporation) and in vitro transcription.
